The ultimate tip I have for lighting is getting a book called "Lighting & Rendering" from Jeremy Birn. Nothing will teach you lighting better than that. If you want to achieve realistic light sets quick and easily, you may try the Python script I wrote a while ago to use HDRShop/LightGen with Poser. With it, you can reproduce natural lighting conditions based on photos in Poser. See the attached link for more on that. What's very important for realism are your surfaces, and I'm not talking about textures here. What makes a difference is how the object's surface reacts to light, reflecting it, absorbing it, transporting it interally... When you hold a sheet of paper against the light, you will see that it looks different than an equally colored and equally thick piece of fabric or plastic. Wet surfaces look different than dry surfaces, polished surfaces look different than dull surfaces and don't forget about fine ripples in most materials. Poser 5's material room will offer you a lot of options here.
