The cinema 4d direct export has different text at the top and throughout." The text is different, but the stuff at the top is just comment lines.. Poser (and other apps) ignore those. The rest of the file might differ as well, but the essense of them should be the same - sets of vertex, texture vertex and/or normal vertex records, followed by facet records, with group and material records spread throughout. In short, no. The file doesn't need to bre filtered through Poser for Poser to understand it. But, it does need to be pre-scaled to the right size and have the groups set up correctly. I usally import a Poser mesh into C4D (like Posette, or V3) with the import scale factor set to '1000' and use that as a guage to get the size right. Then when you export your mesh, use the same '1000' scale factor and it'll load into Poser at the correct size.
