I agree with fixer's method and I'll add a few tips to go with it.
If you can spare the cash, get Correct Reference(from hogsoft.com) and run it BEFORE you remove your existing runtime. This will clean up your texture/object links to all of the cr2s and props in your library.
Another backup method would be to buy another internal harddrive, temporarily install it into your existing computer and copy your runtime to it. Then install this new harddrive into your new machine as a slave drive and install poser onto it. The point here is to have Poser completely isolated from your main harddrive (usually C) which contains the OS. I've learned that Poser tends to run a bit faster this way.
After your new install, do a search here in the forums for tips on setting up virtual memory with Poser. Bear in mind I know next to nothing about Vista, so it's possible that your new computer (with Vista?) may not need any VM tinkering.
