My suggestion would be to just edit your LibraryPrefs.xml file, changing the drive letter for your external runtime folder from E: to S: .
This file is in your user profile, in the same place as your Poser preferences (.ini) file. Look for the lines starting with <ContentFolder folder=.... and ending with .....index="n" >
Don't change the index="0" or index="1" lines. Go to the index="2" line and change the drive letter you see there in the folder name. I can't show a real example because I'm on a Mac, and the paths are different, but it should look something like this:-

<ContentFolder folder="C:Program FilesPoser ProRuntimelibraries" index="0" >
......
<ContentFolder folder="C:Program FilesPoser ProDownloadsRuntimelibraries" index="1" >
