Just make a folder that isn't the system fonts folder and move the ones not being used there. If and when you need them, move them back. Call it something like "MyFonts". If you're clever you could even make a shortcut that references the system fonts folder, so you don't have to dig too much from the MyFonts folder, etc.
I think when people say to "lose the fonts", not to remove it from the computer completely - but rather only the system fonts folder.
But at 10,000+ fonts, you probably need the management software like the one mentioned above. With that many you may also have some redundant fonts (different names, but essentially the same) to weed out as well.
