Windows 7 supports the creating and mounting of virtual hard disks(vhd). Create one and use it to generate the license. The vhd is just a file so you can backup and restore it easily.

In any direct manner, no. Sometimes, you're just not going to have a USB stick available to you, or all of your ports are full with things you need (keyboard, mouse, etc.). Using a VHD with Windows 7 is no different than using a USB stick...for all intents and purposes, that's what a USB stick is; a small hard drive.
HOWEVER, this is not to say that it does not violate their license. I haven't read their license and whether or not it requires that the key be put on a USB stick/dongle or not, so you'll want to contact someone at Messiah to clarify for you, or thoroughly read your license when you receive it.
