Well, if you do reinstall Poser, you won't affect anything inside Poser that you installed after it. I've had to reinstall Poser, and the extra characters, etc. stayed intact. (You would have to reinstall the 4.03 patch, though.) I haven't had the kernel32.dll error so I can't tell you what might be the problem. Hopefully it is not a W ME issue.
