Well, I wish you the best of luck with this. (Bear in mind that large studios like Pixar can spend anywhere from a month up to a year working on a 2 minute sequence).

What I have found works well is the following:
Import your poser object. Leave the "use poser shader tree" and "repose inside vue" options unchecked.
Once you have your object positioned in the scene as you want it, save the scene.
If you need to adjust the pose, go back into poser and adjust it there. When you next open vue, it will detect the change in the poser object and ask if you wish to update it.
When you are finally happy with the pose, export the object as a vob file to save memory. Then, using the new vob file, render your scene.

Not to rain on your parade, but what is your time frame for a project this size? And what resources do you have? Tackling a CG film of that length would require a decent number of people, and then you have to factor in render time. At 30 frames per second at 640 x 480 resolution (or larger if you want HD or film quality), you'd definately have your work cut out for you.
