Enables conformant OpenGL texture clamp behavior.
Texture clamping refers to how texture coordinates are handled when they fall outside the body of the texture. These can be clamped to the edge or within the image.
ON or OFF

and this option should be on or off

Disables driver support for enhanced instructions used by certain CPUs.
Some CPUs support additional 3D instructions that complement your NVIDIA GPU and improve performance in 3D games or applications. This option lets you disable support for these additional 3D instructions in the drivers, which can be useful for performance comparisons or for troubleshooting.
ON or OFF.

Are you using the "Drape from zero pose" option? Does any part of the clothing intersect the figure when in the zero pose? If it does this will result in a failure to calculate the simulation.

And the number crunching is Poser-specific, CPU only. It doesn't matter what your video card is set to; it won't change the speed of the cloth system calculations.

If nothing happens for a long time with the cloth room, it usually does mean that there is some mistake with the cloth setup even if it is just a small mistake like a few polygons of the cloth intersecting with the figures hand.
